    目的:探讨circAff4在体外高糖培养足细胞中的作用,及其与Notch通路及nephrin之间的关系。方法:30 mmol?L-1高糖培养足细胞0、12、24、48、72 h后,采用PCR方法测定circAff4的水平。通过circAff4过表达载体增加circAff4表达后,采用免疫细胞化学、Western blot、PCR方法测定Notch1、Hes1和nephrin的水平。结果:高糖培养足细胞可抑制circAff4表达(P<0.05或P<0.01)。在高糖培养的足细胞中加入circAff4过表达载体增加circAff4表达后,Notch1和Hes1表达明显增加,nephrin表达明显减少(P<0.01)。结论:在高糖培养的足细胞中,circAff4可能通过Notch通路和nephrin参与足细胞损伤作用。

    Abstract:

    Objective: To investigate the role of circAff4 in high glucose-cultured podocytes in vitro and its relationship with Notch pathway and nephrin. Methods: After culturing podocytes in 30mmol?L-1 high glucose for 0, 12, 24, 48, and 72 h, the levels of circAff4 were measured using PCR. Following the overexpression of circAff4 via an overexpression vector, the levels of Notch1, Hes1, and nephrin were assessed using immunocytochemistry, Western blot, and PCR methods. Results: High glucose cultivation of podocytes could inhibit circAff4 expression (P<0.05 or P<0.01). After adding the circAff4 overexpression vector to high glucose-cultured podocytes to increase circAff4 expression, the expression of Notch1 and Hes1 significantly increased, while the expression of nephrin significantly decreased (P<0.01). Conclusion: In high glucose-cultured podocytes, circAff4 may participate in podocyte injury through the Notch pathway and nephrin.
